The objective was to isolate diazotrophic bacteria of the genus Azospirillum spp. and Herbaspirillum spp., from two cultivars of Urochloa brizhanta (Marandu and Xaráes) and one cultivar of Urochloa decumbens (Ipean), test them and evaluate their contribution to the growth of Urochloa brizhanta cv. Marandu identifying the three most promising. This work was carried out at Embrapa Agrobiologia in Seropédica – RJ, in two stages. In the first, isolation, characterization and biochemical tests of diazotrophic bacteria from plants of the Marandu and Xaraés cultivars of U. brizantha and the Ipean cultivar of U. decumbens were carried out in semi-solid culture medium NFb, LGI and JNFb. In this stage, 15 isolates were obtained and were used in the second stage, which consisted of inoculating the 15 isolates plus three standard isolates SP245, ZAE94 and CBAMC in seedlings of U. brizhanta cv. Marandu, plus the uninoculated witness. The results obtained for the dry and fresh weight of the seedlings were tested using the Scott-Knott test at 5% probability and it was possible to select three isolates that were most promising in promoting plant growth. We can conclude that there are nitrogen-fixing, phosphate-solubilizing bacteria that produce 3-indoleacetic acid from Urochloa brizhanta and Urochloa decumbens. Bacteria isolated from cultivars of U. brizhanta and U. decumbens proved to be more efficient in the growth of Marandu grass than standard bacteria from other plant species. Among the 15 isolates tested, isolates J.R.10-4.MB.2*A, L.R.10-3.IB and L.P.10-2.MB differed statistically from the others, being considered the three most promising.
